Polarimeter Calibration Limit

Purpose: To lay down a procedure for calibration of the Polarimeter.

Responsibility: Chemist (s)

Accountability: Head of Q.C. / Q.A.

Procedure: Polarimeter Calibration Limit

1.                 Ensure that Equipment is clean.

2.                 Operate the equipment as per SOP

3.                 Weight accurately 50g sucrose (previously dried) and transfer in 100 ml volumetric flask. Add 50 ml purified water and shake well to dissolve sucrose and make up to 100.0 ml with purified water (solution A).

4.                 Rinse the tube (1-dm) with purified water and filled the tube with purified water after maintaining the temperature 25 ±1ºC.

5.                 Set the null point with the help of knob adjustment and note the reading up to two decimal place with the help of linear & outer side.

6.                 Rinse the tube with sucrose solution (A) and repeat the steps 4 & 5 and measure the reading.

7.                 Similarly prepare solution of 40, 30, 20 and 10g/ 100 ml by diluting 20 ml, 15 ml, 10 ml, 5 ml of solution (A) to 25.0 ml with distilled water and repeat the steps 4 & 5 and note the reading.

8.                 Take five readings for each concentration and then take Average.

9.                 Acceptance criteria:( Polarimeter calibration limit

            10                                                       6.66° ± 0.5

            20                                                       13.30°± 0.5

30                                                       19.93° ± 0.5

40                                                       26.53°± 0.5

                  50                                                       33.11° ± 0.5

Important:

Ø The temperature of solvent & solution should be 25 ± 1ºC.

Ø Polarimeter tube should be filled in such a way to avoid air bubbles.

Ø Solution must be clear.
