SOP on procedure for Glassware cleaning

Purpose
This SOP describes the procedure for Glassware cleaning.
Scope
This procedure is applicable for cleaning of glassware located at ABC Limited.
Procedure :
  • For the cleaning of glassware used for chemical analysis, avoid any chemical residue in glass wares.  Use following cleaning solutions for cleaning of glassware.
  • 0.5 % Teepol or 0.5 % Labolene / Exaline.
  • Chromic acid cleaning solution.
  • Prepare 0.5 % Teepol or Labolene by adding 5 ml Teepol and add it to container containing 995 ml of Purified water and mix well.
  • Use the solution mentioned in section 4.1 for cleaning of laboratory glassware such as bottles, flasks, beakers, and Burettes.
  • Prepare daily fresh cleaning solutions.
  • If glassware becomes dirty or contains coagulated organic matter, clean with chromic acid cleaning solution.
  • Preparation of cleaning solution
  • Weigh 200 g Sodium dichromate and add slowly in a 2 liters container containing 100ml purified water, ice cool it. After cooling add 1500 ml 98% sulfuric acid very carefully.
  • Prepare this solution in a hard borosilicate glass beaker. Use safety glasses during the addition of acid.
  • Maintain the “Cleaning Solution preparation record” refer to Format No. TML/F/RDC/006.
  • Wash glassware as quickly as possible after use but if delays are unavoidable soak the articles in water.
  • Cleaning of glassware, which contains hazardous materials, must be strictly performed by trained person.
  • Use clean hand gloves for cleaning of glass wares.
  • Before cleaning of glassware remove the labeling or marker pen details.
  • Put the used glasswares in the tub containing cleaning solution, for at least 30 minutes before cleaning. After 30 minutes remove the glassware one by one and wash with plenty of potable water and ensure the absence of traces of cleaning solution in the glassware.
  • Finally rinse with purified water.
  • After cleaning, dry the glassware in Hot air oven at temperature not exceeding 60ºC. After drying placed the glassware in appropriate place.
  • When chromic acid solution is used, do not use brush with chromic acid.  When the mixture acquires green color, it should be discarded under continuously flowing water.
  • Most new glassware is slightly alkaline in reaction. For precision chemical tests, new glassware should be soaked several hours in acid water (1% solution hydrochloric acid or nitric acid) before washing.

Transfer Pipettes
Glassware contaminated with water miscible diluents.
  • Place the pipettes after use in a container full of 0.5% v/v Teepol solution for at least 30 minutes. Solution level should be high enough to immerse the maximum portion or all of pipette. Wash the pipettes under the stream of tap water from both ends i.e. dispensing end as well as pipetting end. Take sufficient quantity of raw water in a beaker / reservoir and wash the pipettes by pipetting and dispensing it at least 5 times in such a manner that each pipette shall be washed by 5 times. Finally rinse with purified water. Keep these pipettes in hot air circular oven, temperature maintained at 60 ± 5 °C for 1 hour. After drying, store at clean place.

Glassware contaminated with water immiscible diluents.
  • Wash pipette with IPA and Methanol by pipetting, shaking and dispensing it at least
  • 3 times each. Place the pipettes after use in a container full of 0.5% v/v Teepol solution for at least 30 minutes Wash pipette under the stream of tap water. Take sufficient quantity of water in a beaker / reservoir and wash the pipettes by pipetting and dispensing it at least 5 times in such a manner that each pipette shall be washed by 5 times. Finally rinse with purified water. Keep these pipettes in hot air circular oven, temperature maintained at 60 ± 5 °C for 1 hour. After drying, store at clean place.

Volumetric Flasks
Glassware contaminated with water miscible diluents.
  • Place the Volumetric flask after use in a container full of 0.5% v/v Teepol solution for at least 30 minutes. Take Volumetric flask and wash with raw water by filling it up to about 70% of its Volume, shake it (up & down) and dispense it at least 5 times in such a manner that each flask should  be washed by 5 times. Finally rinse with purified water. Keep these volumetric flasks in hot air circular oven, temperature maintained at 60 ± 5 °C for 1 hour. After drying, store at clean place.

Glassware contaminated with water immiscible diluents.
  • Take volumetric flask after use. Wash the flask with IPA and Methanol by filling it   about 50% of its total volume shake and dispense it at least 2 times.  (Place the Volumetric flask after use in a container full of 0.5% v/v Teepol solution for at least 30 minutes. Wash the flasks with raw water by filling it about 70% of its volume, shake it (up & down)  and dispense it at least 5 times in such a manner that each flask should washed by 5 times, Finally rinse with purified water. Keep these volumetric flasks in hot air circular oven, temperature maintained at 60 ± 5 °C for 1 hour. After drying, store at clean place.

Burette with stopcock.
  • Take the burette after use and half fill it with 0.5% v/v Teepol solution shake and dispense it 2 times, drain the Teepol solution and wash it with raw water, shake it and dispense it at least 5 times in such a manner that each burette should be washed by 5 times. Finally rinse with purified water. And dry the burette by securing in Burette stand in inverted position.

Glass vials used for HPLC & GC analysis.
Glass vials & caps contaminated with water miscible diluents.
  • Take HPLC / GC glass vials and caps after use in a container having 0.5% v/v Teepol solution and sonicate for 10 minutes. Wash thoroughly under the stream of tap water. Place the vials and caps in a container and fill it with water so that all the vials shall be soaked completely. Sonicate it for about 5 minutes and drain the water. Ensure that water shall be drained from vials completely. Repeat the procedure at least two times. Fill the container having same vials and caps with methanol and sonicate for about 10 minutes. Drain the methanol from the container and the vials completely. Rinse the vials and the caps with purified water. Drain the water from container and from vials completely. Keep all the vials and caps in oven, temperature maintained at 60 ± 5 °C for 1 hour. After drying, store at clean place.

Glass vials & caps contaminated with water immiscible diluents.
  • Take HPLC /GC glass vials and caps after use. Place the vials and caps in a container.
  • Fill the container having vials and caps with IPA and shake it for about 5 minutes. Drain the IPA from container & vials completely. Fill the container having vials and caps with methanol sonicate for about 10 minutes. Drain methanol from container & vials completely. Fill the container having vials and caps with 0.5% v/v Teepol solution and sonicate for about 10 minutes. Drain Teepol solution from container & vials completely. Fill the container with water so that all the vials shall be soaked completely. Sonicate it for about 5 minutes and drain the water. Ensure that water shall be drained from vials also. Repeat the procedure at least two times. Finally rinse with purified water. Keep all the vials and caps in oven, temperature maintained at 60 ± 5 °C for 1 hour. After drying, store at clean place.

Glass vials used for Headspace GC analysis.
  • Take GC glass vials after use. Place the vials after use in a container full of 0.5% v/v Teepol solution for at least 30 minutes.  Wash each vial thoroughly under the stream of tap water so that each vial should be free from residue or any deposition. Place the vials in a container and fill it with water. Ensure that all the vials shall be soaked completely. Sonicate it for about 10 minutes and drain the water. Ensure that water shall be drained from vials also. Repeat the procedure at least two times. Finally wash the vials at least 2 times with water and drain it from container and from vials completely. Finally rinse with purified water. Keep all the vials and caps in oven, temperature maintained at 60 ± 5 °C for 1 hour. After drying, store at clean place
