Purpose
The purpose of this SOP is to provide written
guidelines to for operation of
spectrophotometer.
Procedure
- Take out the silica bag from sample compartment.
- Put the mains “ON” by pressing the switch which is on left side of the instrument.
- Initialization will be displayed.
- After initialization will be completed, the eight standard modes will be displayed.
- Press “I” to select photometric mode.
- Press “GO TO WL” key on keyboard. (WL = Wavelength)
- Enter the required wavelength by the numeric keys and press “ENTER”.
- Press “AUTO ZERO” without keeping anything in the sample compartment.
- The absorbance reading will be displayed should be 0.000.
- Clean the cuvettes with distilled water properly.
- Rinse the cuvettes with blank solution 2-3 times & fill it with same solution and wipe out outer surface dry with the help of tissue paper.
- [Note: Cuvette has total 4 surfaces vertically out of which 2 walls are hazy which are exactly facing to each other so at the time of keeping cuvettes in instrument grinded surface to be faced at operating end.]
- Place the blank solution cuvettes in the compartment on the backside. Place the sample solution in the compartment on the front side.
- Reading will be displayed on screen & record it.
Precautions:
Maintain the cleanliness of instrument before &
after use at each time.
Do not allow air bubbles to entrap in path of solution
kept in cuvette as it effects
absorption of light rays.
During analysis stand very close to table to avoid fall
of cuvettes on the floor.
