HPLC Column Washing Method
Procedure:
HPLC column washing procedure for mobile phase
containing buffer:
- After elution of last injection, run the Mobile phase for 10 minutes.
- Flush the injector with sufficient amount of Milli-Q water.
- Run the Milli-Q water (Degassed) through HPLC column by 0.5 ml/min – 1.5ml/min flow where by pressure will not exceed above 3500 psi.
- Flush the column with Milli-Q water for 60 minutes.
- After complete flushing with Milli-Q water, flush the column with solvent like Methanol or Acetonitrile (whatever applicable) for 30 minutes.
- Store the column with end fitting with respective column box.
HPLC Column washing procedure for Mobile Phase (normal
phase):
- After elution of last injection, run the Mobile Phase for 10 minutes
- Flush the injector with sufficient amount of HPLC grade IPA.
- Run the HPLC grade IPA through HPLC column by 0.5ml/min to 1.5-ml/min flow where by pressure will not exceed above 3500 psi.
- Flush the column with HPLC grade IPA for 45 minutes.
- Store the column with end fitting with respective column box.
HPLC column washing procedure for solvent and water
combination phase:
- After elution of last injection, run the Mobile Phase for 10 minutes
- After run the mobile phase, flush the column with solvent like methanol or Acetonitrile (whichever applicable) for 45 minutes.
- Store the column with end fitting with respective column box.
