Purpose
This SOP describes
the procedure for operation and calibration of
UV Cabinet.
Procedure:
Operation
Ensure that the
instrument is free from dust.
Switch on the
mains.
Switch ‘ON’ the
instrument indicator light will be ‘ON’.
Open the cabinet
and place the TLC plate inside and close the cabinet.
Three switches are
provided on the top right corner of the instrument.
Switch
1: For UV-Light at long wavelength (365
nm)
Switch 2: For UV-Light at short wavelength (254 nm)
Switch 3: For Visible
Select
the required UV-Light and switch ON.
Observe
the spot from the TLC plate.
Calibration
(Frequency: Quarterly)
- For the lamp having wavelength (Emission) at about 254 nm:
- Select a pre coated thin layer chromatographic plate coated with silica gel Gas coating substance.
- Prepare a solution of Sodium salicylate in ethanol 95% having a concentration of 0.04%w/v.
- Weight about 40 mg of Sodium Salicylate in 100 ml volumetric flask. Add 20 ml of ethanol 95% to dissolve and make the volume 100 ml with the same.
- Apply
5µl of
this solution on the plate.
- Dry in air and place it in the UV chamber.
- Select the lamp having wavelength (emission) of about 254 nm and check for the compliance.
- The lamp complies if a clear spot of about 5 mm diameter is revealed.
- For the lamp having wavelength (Emission) at about 365 nm:
- Select a pre coated thin layer chromatographic plate coated with silica gel Gas coating substance.
- Prepare a solution of Sodium salicylate in ethanol 95% having a concentration of 0.2%w/v.
- Weight about 200 mg of Sodium Salicylate in 100 ml volumetric flask. Add 20 ml of ethanol 95% to dissolve and make the volume 100 ml with the same.
- Apply
5µl of
this solution on the plate.
- Dry in air and place it in the UV chamber.
- Select the lamp having wavelength (emission) of about 365 nm and Check for the compliance.
- The lamp complies if a clear spot of about 5 mm diameter is revealed.
- The distance between
the lamp and the chromatographic plate under examination used in a
pharmacopoeia test should never exceed the distance used to carry out the above
test.
- Document the activity in Calibration Record
- Report any discrepancy observed during calibration of instrument to ADL – In Charge or his designee for corrective and preventive action.
- ADL – In Charge or his designee informs the defect to Maintenance Department service engineer to rectify the defect. Affix ‘Under Maintenance’ label on the instrument.
- Calibration should be required after maintenance.
